Geboren 1962 in Duisburg. Studium der Theaterwissenschaften und der Neueren Deutschen Literatur, Schauspielausbildung am Zinner-Studio in München. Es folgten Engagements am Residenztheater München, an den Vereinigten Städtischen Bühnen Krefeld und Mönchengladbach, am Stadttheater Bern, am Thalia Theater Hamburg, am Nationaltheater Mannheim, beim Festival Maggio Musicale Florenz, am Berliner Ensemble und am Deutschen Theater Berlin. Von 1997 bis 2009 war er Ensemblemitglied am Schauspiel Köln, wo er anschließend weiter gastierte. Er arbeitete u. a. mit Karin Beier, Johan Simons, Karin Henkel, Alvis Hermanis, Jürgen Gosch, Dušan David Pařízek, Stephan Kimmig, Thomas Langhoff und Michael Thalheimer.
Seit der Spielzeit 2009-10 gehört Markus John zum Ensemble des Deutschen SchauSpielHauses. Für seine Darstellung der Titelrolle in Alice Buddebergs »Ödipus«-Inszenierung erhielt er 2013 den Rolf Mares Preis.
